Le masitinib est à l'étude dans le traitement
de la sclérose latérale amyotrophique (SLA)
45 patients ont été recrutés en phase 2
Les autorités ont accepté la transformation de la phase 2 en phase 3,
avec un recrutement potentiel de 210 patients
AB Science SA (NYSE Euronext - FR0010557264 - AB), société pharmaceutique spécialisée dans la recherche, le développement et la commercialisation d'inhibiteurs de protéines kinases (IPK), annonce le lancement d'un programme de développement clinique du masitinib dans le traitement de la sclérose latérale amyotrophique (SLA).
La SLA est une maladie dégénérative rare qui entraîne une atrophie progressive et une paralysie des muscles volontaires . Il ya environ 30 000 personnes atteintes de SLA dans l'Union Européenne (15 000 aux Etats-Unis ) avec plus de 7 500 nouveaux cas déclarés chaque année (4 500 aux Etats-Unis ). Près de 50 % des patients atteints de SLA meurent dans les 3 ans et 90% décèdent dans les 5 ans.
Le programme de développement clinique du masitinib dans la SLA a commencé par une étude clinique de phase 2, avec 45 patients inclus. Les autorités de santé ont décidé de transformer l'étude de phase 2 en phase 3, avec le recrutement potentiel de 210 patients. La lecture de cette étude est attendue d'ici la fin de 2015.
Il s'agit d'une étude de phase 2/3, prospective, multicentrique, randomisée, en double-aveugle, contrôlée versus placebo, en groupes parallèles, évaluant l'efficacité et la tolérance de masitinib versus placebo dans le traitement de patients souffrant de sclérose latérale amyotrophique (SLA). Le traitement sera administré comme traitement d'appoint aux patients qui ont été traités avec une dose stable de riluzole. L'étude vise à évaluer l'effet du masitinib sur le handicap fonctionnel des patients évalués par la Amyotrophic Lateral Sclerosis Functional Rating Scale (ALSFRS).
Le professeur Olivier Hermine, président du comité scientifique d'AB Science, a expliqué : «Dans cette étude, nous supposons que les mastocytes, cellules immunitaires clés, participent activement à la pathogenèse de la SLA, à travers la libération de médiateurs qui soutiennent le réseau inflammatoire du système nerveux central . Les mastocytes, qui sont présents en grandes quantités dans le cerveau et dans la moelle épinière, peuvent aussi influencer la survie et les fonctions des neurones moteurs, et participer ainsi à la physiopathologie de la SLA. Comme le masitinib est un inhibiteur sélectif de c-Kit et Lyn, deux kinases jouant un rôle majeur dans la survie et l'activation des mastocytes, elle peut conduire à des effets positifs sur les symptômes de la pathologie. "

About ALS
Essential features of ALS are progressive signs and symptoms of lower motor neuron dysfunction (atrophy, cramps, and fasciculations) associated with corticospinal tract signs (spasticity, enhanced and pathological reflexes). Even though the incidence of ALS is similar to that of multiple sclerosis, the prevalence is only 4-6/100,000 (about 25,000 patients in the United States), due to the higher mortality rate. The course is relentless with declines in strength, respiratory function and overall function during the active phase of the disease. No treatment prevents, halts or reverses the disease, although riluzole use is associated with a slight prolongation of survival.
About masitinib
Masitinib is a new orally administered tyrosine kinase inhibitor that targets mast cells, important cells for immunity, as well as a limited number of kinases that play key roles in various cancers. Owing to its novel mechanism of action, masitinib can be developed in a large number of conditions in oncology, in inflammatory diseases, and in certain diseases of the central nervous system. Through its activity of inhibiting certain kinases that are essential in some oncogenic processes, masitinib may have an effect on tumor regression, alone or in combination with chemotherapy.
